Effect of the Internal Septum Extract of the Walnut Kernel on the Mesenchymal Stem Cells Cycle and MSCs-derived Insulin-Producing Β-cells Differentiation and Glucose Uptake

Background: For years, scientists have been striving to identify compounds improve the characteristics and functions of cells for therapeutic applications. Objectives: This study aimed evaluate effect aqueous decoction internal septum walnut kernel (DISWK) on survival, total antioxidant capacity (TAC), cell cycle rat bone marrow-derived mesenchymal stem (rBM-MSCs). We also evaluated expression genes involved in pancreatic insulin-producing β-cell (IPCs) commitment glucose uptake MSCs-derived IPCs. Methods: The rBM-MSCs were isolated, then cytotoxic effects DISWK examined by MTT TAC assays, respectively. Moreover, influence was PI-staining flow-cytometry analysis. Next, transdifferentiated into IPCs treated with DISWK. Finally, Ins1/2, Insr, Glut1, Glut4, Pdx1 investigated DISWK-treated qRT-PCR. Results: results indicated that significantly reduced viability a dose- time-dependent manner decreased their long-term cultures. it suppressed at S G2 phases. gene analysis showed upregulated Glut1 downregulated rBM-MSCs-derived Conclusions: Decoction suppresses MSCs may prevent development via inhibiting Pdx1.
